Leukocyte differential test is a widely carried out clinical procedure for screening infectious diseases. Existing hematology analyzers require labor-intensive work and panel of expensive reagents. Herein, an artificial-intelligence-enabled reagent-free imaging analyzer (AIRFIHA) modality reported that can accurately classify subpopulations leukocytes with minimal sample preparation. AIRFIHA realized through training two-step residual neural network using label-free images isolated acquired from custom-built quantitative phase microscope. By leveraging the rich information contained in images, not only high accuracy achieved differentiating B T lymphocytes, but also CD4 CD8 cells are classified, therefore outperforming classification most current analyzers.
